DESCRIPTION
Cyanine3 SH is a sulfhydryl-reactive Cy3 derivative featuring a terminal thiol (-SH) group for site-specific conjugation to maleimide, iodoacetyl, or pyridyldithiol functionalized molecules. This non-sulfonated reagent enables controlled labeling of proteins, peptides, and surfaces while preserving the photostability and brightness characteristic of Cy3 fluorophores.
The thiol group (pKa ~8.3) facilitates:
• Maleimide coupling (pH 6.5-7.5, 1-2 hr at 25°C)
• Disulfide exchange (pH 8.0 with TCEP reduction)
• Gold surface immobilization via Au-S bonds
